Paul O'Connor, who played with the Kilmurry Ibrickane club until last year, will line out at centre-back at Gaelic Park.
Mayo will start 12 of the 15 players who lined out in last year's All-Ireland Final.
Elsewhere, All-Ireland winner Mark McHugh has left the Donegal panel.
Selector Damien Diver says the player has been suffering with injuries and indifferent form and needed some time out.
Diver said that the door is open to a return for Mc Hugh.
Thomas Mc Kinley, Gary McFadden and Antoine McFadden have also left the squad.
